How Nitrolingual Affects PatientsNitroglycerin relaxes the walls of your blood vessels, allowing for increased blood flow to the heart and a decrease in the pressure of your heart's walls. This can help reduce angina symptoms. However, taking nitroglycerin for a long time can cause your body to become used to it, resulting in it becoming less effective. It may also cause problems like increased sensitivity to other substances that make your blood vessels constrict.
How Nitrolingual works in the bodyNitroglycerin works to widen your blood vessels, allowing for easier blood flow. It does this by converting to nitric oxide in smooth muscle cells. Nitric oxide activates an enzyme called guanylate cyclase, which in turn produces a compound called cyclic guanosine 3',5'-monophosphate (cGMP) in the walls of your veins, arteries, and cardiac tissue. This compound causes protein kinase-dependent phosphorylation and leads to your blood vessels relaxing, allowing for increased blood flow.

Nitrolingual Toxicity & Overdose RiskThe toxic dose of nitroglycerin for rats is 105 mg/kg when taken orally and 23.2 mg/kg when taken intravenously. Overdosing on nitroglycerin can cause severe low blood pressure, headache, dizziness, rapid heartbeat, skin flushing, nausea, vomiting, shortness of breath, confusion, and paralysis. In rare cases, a condition called methemoglobinemia may occur, which can be treated with methylene blue unless a person has a known deficiency in an enzyme called G6PD. There is no known antidote for nitroglycerin overdose, so treatment involves managing the symptoms
