How Pyrazinamide Affects Patients

Pyrazinamide is a medication used to fight tuberculosis. It only works against the bacterium Mycobacterium tuberculosis. Pyrazinamide is activated in the bacterium to Pyrazinoic acid, which stops the bacteria from making new fatty acids needed for growth and replication.

How Pyrazinamide works in the body

Pyrazinamide works by collecting inside of active _M. tuberculosis_ cells when they express the pyrazinamidase enzyme. It accumulates in acid conditions and is thought to work by preventing the bacterium from producing fatty acids, disrupting the cell's membrane potential, and inhibiting trans-translation. These effects prevent the cells from reproducing, causing them to die.

When to interrupt dosage

The dosage of Pyrazinamide is contingent upon the diagnosed state, such as Active Tuberculosis, antituberculosis treatment and Pulmonary Tuberculosis (TB). The measure of dosage likewise depends on the method of delivery, as detailed in the table below.

Patient Q&A Section about pyrazinamide

These questions and answers are submitted by anonymous patients, and have not been verified by our internal team.

What type of antibiotic is pyrazinamide?

"This medicine is used to treat the symptoms of tuberculosis and may be used alone or with other medications. It belongs to a class of drugs called antitubercular agents."

Answered by AI

How long do you take pyrazinamide for TB?

"The bacteria that cause tuberculosis are difficult to eliminate. Your child will need to take up to four medications for at least three to six months. Pyrazinamide is one of these vital medications. It is given for the first two months of treatment."

Answered by AI

What is the side effect of pyrazinamide?

"You may experience nausea, vomiting, loss of appetite, or mild muscle/joint pain. If any of these effects persist or worsen, tell your doctor or pharmacist promptly."

Answered by AI

What is the mechanism of action of pyrazinamide?

"The mechanism of action for PZA is unknown. The parent compound enters the bacterium passively and is metabolized via pyrazinamidase (PZase) within the cytoplasm to pyrazinoic acid; pyrazinoic acid is the active form of the drug."
