Methotrexate Toxicity & Overdose RiskThe lowest toxic dose of leucovorin in rats has been found to be 135mg/kg and 146mg/kg in mice. Signs of overdose include a decrease in white blood cells, platelets, and red blood cells; mouth sores; nausea and vomiting; stomach ulcers; and bleeding from the digestive tract. In case of an overdose, patients should be treated with glucarpidase, and should not be given leucovorin for 2 hours before or after receiving glucarpidase.

12/6/2017 I started taking this medication seven weeks ago at a dosage of 10mg per day. After four weeks, I increased my dosage to 12.5mg per day as instructed by my doctor. I have seen mixed results so far; some of my plaques are clearing up gradually, but the larger ones don't seem to be affected much yet. I haven't experienced any major side effects other than some abdominal discomfort. Additionally, while my joint pain and stiffness has not worsened (as it usually does during the winter months), neither has the swelling gone down or my joints become more functional (they're only at about 90% right now).

10/4/2017 I've been taking this once-a-week pill for two years now with no adverse effects. That being said, it hasn't completely stopped the inflammation caused by my psoriatic arthritis--I still have swollen and painful joints in my hands and feet. My doctor also prescribed Meloxicam for pain control, as well as D3, folic acid, and fish oil.
