How Indocin Affects PatientsIndometacin is a type of painkiller and fever reducer used to treat inflammation in rheumatoid arthritis. It works by reducing pain, fever, swelling, and stiffness in the joints. It has been used to treat gouty arthritis, but it can also increase your risk of serious heart problems, like a stroke or heart attack. Additionally, it may lead to stomach bleeding, ulcers, or perforations. Studies have also suggested that indometacin may reduce blood flow to the brain, although the significance of this is not clear. It may also inhibit neuron activity in certain areas of the brain.
How Indocin works in the bodyIndometacin works by blocking two enzymes, cyclo-oxygenase and phospholipase A2. The enzymes normally convert substances in the body into prostaglandins, which help create inflammation. By blocking these enzymes, indometacin reduces inflammation and pain. It can also reduce fever by increasing blood flow to the skin and helping the body release heat. Indocin Toxicity & Overdose RiskThe toxic dose of indomethacin for rats is 2.42mg/kg and 13mg/kg for mice. Signs of an overdose include nausea, vomiting, headache, dizziness, confusion, or fatigue. There have also been reports of tingling, numbness, or seizures. If an overdose occurs, the doctor may induce vomiting or pump the stomach to clear the drug. The patient should then be monitored for signs of stomach ulcers or bleeding, and antacids may be used.

Indocin for Gout
Indocin was a total life-saver for me. I had debilitating hip pain that no one could figure out, until my MD did an exam and determined it might be joint related. He ordered an MRI, but in the meantime prescribed Indocin. Within two hours of taking the first dose, my pain completely subsided. I've been on the prescription regimen for two days now with zero side effects and zero pain.

I'm currently taking this medication to help with the pain of my intracranial hypertension (or pseudotumor cerebri), however since starting this medication my headaches have actually gotten worse. Has anyone else had a similar experience? If so, I may stop taking this altogether.

Indocin for Rheumatoid Arthritis
I was prescribed this for an extremely painful ankle, which the doctor said was likely gout. He said it would help the pain quickly, but 12 hours later I'm still in agony. A shorter dose of a stronger pain medication would have been much better than this.

Indocin for Gout
I had a severe outbreak of gout and my doctor prescribed Indocin. You must take this drug with food, or else you'll get dizzy and likely have diarrhea. I've only taken two pills so far but it has definitely reduced the pain and swelling. Another great thing for reducing swelling is Turmeric; it's a natural herb that works well with no side effects!

Indocin for Inflammation of the Tendon
I was prescribed Indocin for my chronic headaches, but it only marginally helped me over the course of six weeks. I also think that doctors should be more careful about prescribing this medication to patients with hypertension; my blood pressure skyrocketed while taking it and I ended up in the ER after passing out while using a snow blower. For now, I've decided to stop taking Indocin until my doctor can re-evaluate it.

I had cellulitis/phlebitis from an insect bite and Indocin did help reduce the swelling, but I couldn't continue use because of the diarrhea and stomach cramps it caused.

Indocin for Inflammation of the Lining of a Joint
This treatment has helped me with my chronic gout, but this morning I took it without food and within an hour felt lightheaded and faint. This made me very anxious, but after doing some research online I found out that this is a common side effect of the drug. I also experienced diarrhea, but the alternative is that I can't walk, so I guess you have to pick your poison.

Ibuprofen for Patent Ductus Arteriosus

Newborns born early are at risk for a serious health problem called patent ductus arteriosus (PDA). PDA is a passageway between heart and lung that can cause life-threatening complications such as bleeding in the brain or even death if it remains open and large. When closure of PDA is needed, doctors make every attempt to do it as soon as possible. Ibuprofen is the best drug to close the PDA, but it only works for 50% of small newborns. The investigators have shown before that small newborns handle ibuprofen differently and the amount of active ibuprofen that reaches their blood can be very unpredictable. Studies have shown if enough ibuprofen reaches the body, it can close the PDA. Therefore the investigators designed this study to see whether it is possible to give each newborn the right amount of ibuprofen that their body needs to close the PDA. The investigators will compare two ways to give ibuprofen in a small number of newborns: 1 - standard amount of ibuprofen to everyone, which is the usual care or 2 - ibuprofen doses that will be changed based on how much active ibuprofen has reached the body and how well the newborn's PDA is closing. The investigators will then compare the number of PDAs closed in each group and closely monitor any possible challenges for this new practice. By doing this project, the goals can be summarized as below: A. Primary goal: To determine if it is feasible to successfully run a larger study in the future. B. Secondary goals 1. To assess how well and how safely the personalized (MIPD) method works, using a tool called WAPPS-PDA to guide dosing. 2. To compare the effectiveness and safety of the personalized method with standard ibuprofen dosing. 3. To identify drug levels in the blood (Cmin, AUC0-24, AUC0-72) that are associated with complete, partial, or no response to treatment.

Temporo-masseteric Nerve Block for Postoperative Pain

The proposed design is a triple-blind, placebo-controlled randomized prospective phase II trial to assess the feasibility of employing the Temporo-masseteric Nerve Block (TMNB) injection to decrease jaw muscle pain, overall pain, improve mouth opening and minimize pain medication consumption. 80 participants undergoing lower third molar removal under sedation will be enrolled. Those who develop significant jaw muscle pain on the following day will be randomized to receive either the TMNB injection or a placebo. Daily ecological momentary assessments (EMAs) will be collected reflecting elements of patient experience for all participants. On Day 8, participants return for an end-of-study evaluation and return their unused pain medications. Up to 40 participants who are randomized to receive TMNB/placebo will be recruited to participate in a qualitative interview regarding their experience.
