Treatment Summary
Doxepin is a medication used to treat anxiety and depression. It works by blocking certain receptors in the brain to reduce feelings of anxiety and depression. It is similar to other medications in its class, such as amitriptyline, clomipramine, desipramine, imipramine, nortriptyline, protriptyline, and trimipramine. Doxepin was originally developed by Pfizer and approved by the FDA in 1969 as an antidepressant, but it was later approved in 2010 for the treatment of insomnia.

How Doxepin Hydrochloride Affects Patients
Doxepin is a type of antidepressant that is known to cause antidepressant, sedative, and anticholinergic effects. It works by decreasing the electrical activity of the brain and is usually prescribed to treat insomnia or anxiety. The antidepressant effects start to take effect after two weeks, while the sedative effects are felt right away. High doses of doxepin can cause central nervous system depression and limit its effectiveness due to its antiadrenergic and anticholinergic properties.
How Doxepin Hydrochloride works in the body
Doxepin works by blocking histamine receptors and increasing the amount of certain chemicals in the brain. This helps to reduce anxiety and depression by increasing activity at the nerve cell receptor sites, desensitizing serotonin and beta-adrenergic receptors, and increasing dopamine transmission in the frontal cortex.
When to interrupt dosage
The suggested dosage of Doxepin Hydrochloride is contingent upon the diagnosed condition, such as Depression, Involutional, Mental Disorder, Organic and Insomnia. The amount of dosage likewise deviates as per the approach of delivery (e.g. Oral or Tablet - Oral) outlined in the table beneath.

Doxepin Hydrochloride Toxicity & Overdose Risk
The highest toxic dose of doxepin in mice and rats has been found to be 180mg/kg and 147mg/kg, respectively. Overdosing on doxepin can cause seizures, irregular heart rhythms, loss of consciousness, low blood pressure, and depression of the central nervous system. Studies have also shown that doxepin can affect fertility, causing an increase in the copulatory interval, decrease in the number of viable embryos, and reduction in sperm motility. There is no evidence suggesting doxepin can cause cancer or mutations.
