Treatment Summary
Zelnorm is a medication developed by Novartis that was first approved by the FDA in 2002 to treat irritable bowel syndrome with constipation (IBS-C). In 2007, due to concerns of potential cardiovascular risks, the drug was removed from the US market, but was re-approved in 2019 for female patients under 65 who are at a lower risk of a cardiovascular event. Zelnorm works by targeting serotonin-4 (5-HT(4)) receptors in the gut wall and smooth muscle cells to help with esophageal relaxation, peristaltic gut movement, and natural secretions. However, it is still not

How Zelnorm Affects Patients
Tegaserod works by activating serotonin type-4 (5-HT(4)) receptors and blocking 5-HT(2B) receptors. It has no effect on 5-HT(1), 5-HT(3) or dopamine receptors. In clinical studies, the drug did not cause a dangerous increase in heart rate, though it did lead to an increase of up to 60 milliseconds in 7% of patients. The same studies showed that tegaserod and its main metabolite could increase platelet aggregation in laboratory tests, but it is unclear what this means for people taking the drug.
How Zelnorm works in the body
Irritable bowel syndrome (IBS) is a condition that causes abdominal pain, bloating, and other uncomfortable issues with the digestive system. One potential way to treat IBS-C (constipation-predominant IBS) is through 5-hydroxytryptamine (5-HT or serotonin). 5-HT can cause the release of chemicals in the gut that help move material through it, as well as relax the esophagus and reduce muscle contraction in the colon. It also helps to create natural fluid secretion in the gut. Tegaserod is a medication that works by activating 5-HT(4) receptors

Zelnorm Toxicity & Overdose Risk
Taking more than the recommended dose of Tegaserod can cause diarrhea, headaches, abdominal pain, flatulence, nausea, and vomiting. Pregnant women should be cautious when taking Tegaserod, as animal studies have shown decreased body weight, delays in developmental landmarks, and decreased survival in rat pups. Breastfeeding is not recommended while taking Tegaserod due to the potential for serious reactions in the breastfed infant. The safety and effectiveness of Tegaserod in pediatric patients and those over the age of 65 has not been established. In animal studies, there was no evidence of carcinogenicity at lower doses but mucosal hyperpl
