How Ambrisentan Affects PatientsAmbrisentan is a drug that is generally taken once a day. Taking 10 mg of ambrisentan does not change the heartbeat, but taking 40 mg can cause a slight increase. It does not cause major changes to the heartbeat without other medicines. Taking ambrisentan for 12 weeks has been seen to decrease a certain hormone in the blood (BNP) by 29-45%, while taking a placebo caused an 11% increase.
How Ambrisentan works in the bodyEndothelin-1 (ET-1) is a natural body molecule which can cause increased pressure in the lungs in some people. Ambrisentan works by blocking ET-1 from attaching to its receptors, stopping it from causing the blood vessels to constrict. This allows the vessels to stay wide open, reducing pressure and improving lung function. Ambrisentan only blocks the action of ET-1, not other molecules like nitric oxide and prostacyclin, so it doesn't interfere with the body's natural processes.
