Estradiol is a hormone naturally found in females. It is used in hormone therapy to treat conditions caused by low estrogen levels, such as hot flashes and vaginal dryness. Estradiol can be taken orally, injected, used as a vaginal ring, patch, spray, gel, or cream. To make it more suitable for oral use, estradiol is usually combined with an ester side-chain, such as ethinyl estradiol (EE). Estradiol is used to reduce symptoms such as hot flashes and vaginal dryness. It has also been shown to increase bone density and make changes to the body's lipid profile. Taking estradiol can also increase the levels of certain proteins in the liver, such as sex hormone binding globulin and thyroid-binding globulin, and suppress the production of follicle-stimulating hormone in the pituitary gland. How Vivelle works in the body
Estrogen is found in many parts of the body, including the brain, skin, and reproductive organs. In women who are still menstruating, estrogen is mainly produced by the ovaries. After menopause, the main source of estrogen is the adrenal glands. Estrogen works by attaching to two types of receptors, ERα and ERβ, as well as a receptor called GPER. Once attached, it enters the nucleus of the target cell and changes how certain genes are expressed. This results in certain effects, such as reducing symptoms like hot flashes, chills, mood swings, and dryness in postmenopausal women.

Studies in rats have found that taking estradiol orally over a 90 day period can cause blood, female reproductive, male reproductive, endocrine, and liver toxicity when taken in doses of up to 0.003 mg/kg per day. The highest dose of estradiol that has been tested on humans is 21 mg/kg taken intermittently over 21 days. The highest toxic dose of estradiol in rats is 960 mg/kg.
