Isotretinoin is a derivative of vitamin A used to treat severe acne. It was previously sold under the brand name Accutane, but is no longer available. Isotretinoin has significant risks during pregnancy, which is why it is only available through the iPLEDGE program in the United States. This medication was approved by the FDA in 1982.

The way isotretinoin works in the body is not well understood.

How Sotret works in the body

Isotretinoin works in several different ways to reduce acne. It decreases the amount of oil produced in the skin, reducing blockages in the pores and making it harder for bacteria to grow. It also causes cells in the skin to die, and reduces the size of pores. Isotretinoin also makes it harder for bacteria to survive by making the environment less hospitable. It may even boost the immune system and reduce inflammation. Finally, there is evidence that Isotretinoin interacts with FoxO1, which could help explain its other effects.

The suggested dose of Sotret is dependent upon the declared condition, such as Acne conglobata, Acne Vulgaris and Pregnancy. The measure of dosage is contingent upon the modality of delivery (e.g. Capsule - Oral or Capsule) featured in the table beneath.

Sotret Toxicity & Overdose Risk

Symptoms of an isotretinoin overdose may include vomiting, redness on the face, sores in the mouth, abdominal pain, headache, dizziness, and difficulty with coordination. The lowest dose causing toxic effects for children is 30mg/kg/21W, 24mg/kg/4W in men, and 56mg/kg/8W in women. In animal studies, doses higher than the recommended clinical dose have been linked to an increased risk of tumors of the adrenal gland and testicular atrophy. I had tried everything else and this was my last hope. It was a bit of a process to get started (birth control, pregnancy and blood tests, signing into the program...) but it was well worth it. I am in my second month of treatment now and have not had any new acne since my first 2 weeks on the medicine. Furthermore, the redness from my previous acne bumps are beginning to fade.

I've now been on this medication for four months, which is one month longer than my doctor said should be necessary. I have seen a significant improvement, but I worry about the other 15% of people who don't see complete results.

The treatment I received was great and it finally cured my acne. I was recommended to take a multivitamin supplement without Vitamin A since the Sotret medication is related to Vitamin A and taking the two together can result in potential side effects.

This has been great! I've tried every type of oral medication you could imagine for my face and this is the only one ive seen improvement in. I've only been taking this for less than a month and almost EVERYTHING is gone. Despite the dry lips, peeling skin, and noticable mood changes this is definitely worth it. If nothing has worked for you yet definitely try this, you won't regret it!

My son used this medication as directed, and while it did clear up his acne, he developed severe depression that he had not experienced before. Unfortunately, three months later he committed suicide. I am devastated by this turn of events and believe that using this medication was the wrong decision.

I highly recommend this drug for anyone struggling with acne. It's working really well for me and is much easier than other treatments I've tried in the past. The only downside is that it can cause your skin to dry out, as well as your nose.

I've been struggling with a skin issue on my face for the last few months. It's red, swollen, and itchy, and no matter what I do, I can't seem to get rid of it. My doctor prescribed Sotret, but even after four months of treatment, there's been no improvement. If anything, it seems to be getting worse. Not sure if I should continue taking this medicine or not.

My son started taking this when he was 17. We had tried everything for more than 2 years and nothing worked. But after 6 months on this medication, he was a completely different person. It did wonders for him inside and out. I would tell all parents to try it for their kids' sake, but just be mindful of the potential side effects.

I started taking this drug in 2005 to treat my acne. It did improve my skin, but since then I've noticed increased hair loss and thinner hair overall. I also seem to get food poisoning more often than before.

Talimogene Laherparepvec + Nivolumab for Skin Cancer and Lymphoma

This phase II trial studies how well talimogene laherparepvec and nivolumab work in treating patients with lymphomas that do not responded to treatment (refractory) or non-melanoma skin cancers that have spread to other places in the body (advanced) or do not responded to treatment. Biological therapies, such as talimogene laherparepvec, use substances made from living organisms that may stimulate or suppress the immune system in different ways and stop tumor cells from growing. Immunotherapy with monoclonal antibodies, such as nivolumab, may help the body's immune system attack the cancer, and may interfere with the ability of tumor cells to grow and spread. Giving talimogene laherparepvec and nivolumab may work better compared to usual treatments in treating patients with lymphomas or non-melanoma skin cancers.
