How Taclonex Affects PatientsCorticosteroids are a type of medicine that reduce inflammation and inhibit pro-inflammatory signals. Depending on the patient, a dose much larger than normal may be prescribed. People who need to take a corticosteroid for a long time should be aware of the risk of the body not producing hormones correctly and the increased likelihood of getting infections.
How Taclonex works in the bodyGlucocorticoids like betamethasone work by reducing inflammation and encouraging anti-inflammatory reactions. They block molecules that trigger inflammatory responses, such as NF-Kappa B and other transcription factors. They also stop the formation of arachidonic acid derivatives. Betamethasone works in two ways: it either activates receptors to start a slower, anti-inflammatory process, or it uses existing receptors to quickly reduce current inflammation.

Taclonex Toxicity & Overdose RiskTaking high amounts of glucocorticoids for a long period of time can cause cataracts, glaucoma, high blood pressure, water retention, high cholesterol, stomach ulcers, pancreatitis, muscle weakness, mood changes, psychosis, thinning skin, allergies, acne, excessive hair growth, reduced immunity, diabetes, low calcium and phosphorus levels, acid buildup in the body, slowed growth in children, and decreased hormone production in the adrenal glands. Treatment for overdose includes reducing the dose or stopping the steroid, as well as providing symptom relief and supportive care.

After trying cortisone and other topical treatments without success, I was given a sample of taclonex by my dermatologist. This completely cleared up my issue within three weeks. Now, if I get the occasional spot, I use it a couple times a month. Even with a coupon, though, my copay is still $55.

This gel form of the drug is just as effective as anything else I've used in the past. The main issue I have is that it's difficult to tell how much is actually coming out and if any is being wasted. With liquid drugs, it's easier to feel them come out.

A few years ago, I used this treatment on my feet. It did help my feet, but now I have developed painful red patches on my palms and fingers which peel. I believe that this product has permanently thinned my skin, and I am considering legal action. Do not use it.

ESK-001 for Plaque Psoriasis

The objective of the ESK-001-018 long term extension is to evaluate the safety and efficacy of ESK-001 over time. The scientific questions it aims to answer are: * How safe is taking ESK-001 long-term in people with moderate to severe plaque psoriasis? * Does taking ESK-001 long-term reduce the severity of people's plaque psoriasis? Patients will enter the long-term extension study following completion of one of the parent studies (ESK-001-016 or ESK-001-017) and will receive open-label ESK-001 twice daily for 24 weeks. After 24 weeks, the first 200 patients meeting at least PASI-75 clinical response will be randomly assigned to receive ESK-001 or placebo. At any point during this time, the patients losing the initial clinical response may return to the open-label ESK-001 treatment. Patients who complete Week 48 will return to open-label ESK-001 treatment and they will receive ESK-001 until the end of the study or discontinuation. All the remaining patients not meeting the entry criteria for the randomized withdrawal phase will continue to receive open-label ESK-001 for the remainder of the study. Patients taking part in the study must be men or women aged at least 18 years old and have completed a previous (parent) study of ESK-001 in moderate to severe plaque psoriasis. Patients must consent and agree to: * ensure drug daily compliance until end of study or discontinuation. * visit the clinic for checkups and assessments. * provide blood and urine samples.

The goal of this clinical trial is to learn if IRX4204 works to treat plaque psoriasis in adults. It will also learn about the safety of IRX4204. The main questions it aims to answer are: * Does IRX4204 treat plaque psoriasis symptoms? * Does IRX4204 treat plaque psoriasis symptoms better than someone who is not being treated? * What medical problems do participants have when taking IRX4204? Researchers will compare IRX4204 to a placebo (a look-alike substance that contains no drug) to see if the drug works to treat mild to moderate plaque psoriasis. Participants will: * Take IRX4204 every day for 28 days * Visit the clinic once every week for checkups and tests * Complete specific assessments about plaque psoriasis and changes to plaques
