What is Janumet
Sitagliptin
The Generic name of this drug
Treatment Summary
Sitagliptin is a diabetes medication that works by lowering blood sugar levels. It is usually taken in combination with diet and exercise, and was approved by the FDA in 2006. The drug works by blocking an enzyme called dipeptidyl peptidase-4 (DPP-4) that breaks down a hormone called incretin, which helps the body produce insulin and reduces glucagon production. This helps to regulate blood sugar levels in patients with type 2 diabetes.

Effectiveness
How Janumet Affects Patients
Sitagliptin stops an enzyme (DPP-4) from working, leading to higher levels of two hormones (GLP-1 and GIP), lower levels of another hormone (glucagon), and stronger insulin production when exposed to glucose.
How Janumet works in the body
Sitagliptin works by slowing down the activity of an enzyme called DPP-4. This enzyme breaks down a type of hormone called incretins, which help control blood sugar levels. When DPP-4 is inhibited, more incretins are available, which causes the body to produce more insulin and less glucagon. The result is better control of blood glucose levels, as demonstrated by reduced levels of glycosylated hemoglobin (HbA1c).

Janumet Toxicity & Overdose Risk
Studies have not found any negative effects on mothers or babies when taking Sitagliptin at normal doses. However, high doses of the drug have been linked to malformations in ribs in animals. It is not known if Sitagliptin would be expressed in human breast milk. Prescribing the drug should be done cautiously, considering both the benefits and risks. Currently, there is not enough data to determine the safety and effectiveness of Sitagliptin in children. Elderly patients may respond differently than younger patients, so caution should be used when prescribing it to this group. Additionally, Sitagliptin has been linked to an
