Effectiveness

How Aloprim Affects PatientsAllopurinol works by blocking the body's production of uric acid, which is the cause of gout symptoms like painful tophi, joint pain, inflammation, redness, reduced range of motion, and swelling.
How Aloprim works in the bodyAllopurinol works to reduce gout symptoms by blocking an enzyme that creates uric acid. The blocked enzyme then reuses hypoxanthine and xanthine for creating nucleotides and nucleic acid. This reduces the amount of uric acid in your blood, which helps reduce gout symptoms. Allopurinol also increases the amount of hypoxanthine and xanthine in your urine, but these substances are more soluble and are quickly taken away by the kidneys, so they won't form crystals in your body. As a result, allopurinol can help dissolve gout tophi.

The serum concentration of Didanosine can be increased when it is combined with Allopurinol.
Oteracil
Major
The therapeutic efficacy of Oteracil can be decreased when used in combination with Allopurinol.
Pegloticase
Major
The risk or severity of adverse effects can be increased when Allopurinol is combined with Pegloticase.
Abacavir
Minor

Minor
Allopurinol may decrease the excretion rate of Aclidinium which could result in a higher serum level.
Aloprim Toxicity & Overdose RiskThe lowest toxic dose of allopurinol in rats is 10mg/kg, and in mice is 78mg/kg. Studies in animals have found that taking up to 20 times the normal human dose (about 5mg/kg per day) does not impair fertility or cause harm to the fetus. There was one published report of a study in pregnant mice that was given 50 or 100mg/kg of allopurinol, which resulted in increased numbers of dead fetuses and malformations in the offspring. As there are no adequate studies of allopurinol in pregnant women, it should only be taken if
