This study is a first-in-human (FIH), Phase 1a/1b study of BG-68501, a cyclin-dependent kinase-2 inhibitor (CDK2i), to assess the safety, tolerability, pharmacokinetics (PK), pharmacodynamics, and preliminary antitumor activity of BG-68501 in participants with advanced, nonresectable, or metastatic solid tumors as monotherapy and in combination with fulvestrant with or without BGB-43395, a selective CDK4 inhibitor, in adults with hormone receptor positive (HR+)/human epidermal growth factor receptor 2 negative (HER2-) breast cancer (BC). The study will also identify a recommended dose for expansion (RDFE) for BG-68501 as monotherapy and in combination for subsequent disease directed studies.

The study will be conducted in 2 parts: Part 1 (dose escalation and safety expansion) and Part 2 (dose expansion).

I have had a stem cell or organ transplant in the past.

My cancer is advanced or has spread and may respond to treatments targeting CDK2.

I have had hepatitis B or currently have hepatitis C.

My cancer is advanced or has spread, and it is either HR+/HER2-, ovarian, or small cell lung cancer.

I am fully active or can carry out light work.

I haven't had major surgery in the last 4 weeks.

I have been treated with drugs targeting CDK4/6 for my cancer.

I do not have untreated brain metastasis or leptomeningeal disease.

I haven't had any cancer other than the one being studied or non-serious skin cancer in the last 3 years.

I haven't taken antibiotics, antifungals, or antivirals in the last 28 days and don't have COVID-19 symptoms.

I am a woman with advanced breast cancer and am either postmenopausal or receiving hormone therapy to suppress ovarian function.

I've tried all standard treatments for my condition without success or couldn't tolerate them.

My cancer is advanced, has specific gene mutations, and is not HER2 positive.

The purpose of the study is to test the safety and dosing of \[177Lu\]Lu-FF58, a radioligand therapy for patients with advanced or metastatic tumors that express proteins known as integrins: alpha-v beta-3 integrin (αvβ3) and alpha-v beta-5 integrin (αvβ5). The study will also further characterize the radioligand imaging agent \[68Ga\]Ga-FF58 including its ability to identify tumor lesions and its safety profile.

The purpose of this study is to find out whether combining nivolumab, FOLFOX, and regorafenib may be a safe and effective treatment for people who have HER2-negative metastatic esophagogastric cancer.

Nivolumab is an antibody, like the proteins made by the immune system to protect the body from harm. Nivolumab blocks the protein PD-1 (programmed cell death receptor-1) that usually acts as a "brake" on the immune system. Blocking this protein is like releasing the brakes, so that the immune system can target cancer cells and destroy them.

FOLFOX is a combination of three standard chemotherapy drugs (leucovorin, 5-fluorouracil, and oxaliplatin) commonly used to treat your type of cancer. The drugs work by damaging the DNA in cancer cells, which can cause the cells to stop growing and die.

Regorafenib is a type of drug called a tyrosine kinase inhibitor (TKI). This drug targets the tyrosine kinase protein found in or on the surface of cancer cells that the cells need to survive and grow. Blocking this protein may stop cancer cells from growing, or cause them to grow more slowly or to shrink.

The study researchers think that combining nivolumab, FOLFOX, and regorafenib may be a more effective treatment for HER2-negative metastatic esophagogastric cancer than the usual chemotherapy treatment(s) alone.

You have already received treatment with regorafenib.

You have vitiligo or alopecia (hair loss).

You have cancer that has spread to your brain or the lining around your brain.

You have already received treatment with a specific kind of medication that targets proteins called PD-1, PD-L1, or PD-L2.

You have a lung condition called pneumonitis that is not caused by an infection and is currently active.

You have underactive thyroid or a condition called Hashimoto syndrome, but it is being treated with hormone replacement therapy.
